Scotch-Irish and French Huguenots first settled the area in the 18th century. Williamsburg County was founded in 1804 after previously belonging to the Georgetown District. The Revolutionary War battles of Black Mingo, Mount Hope Swamp, and Lower Bridge all took place in the region.

The region first became wealthy from the cultivation of Indigo, and today it remains largely agricultural. It is also known for its nature preserves and outdoor sports.
